扁桃がん (TC) は中咽頭がんの中で最もよく見られるがんであり、その大半は扁平上皮がん (squamous cell carcinomas) である。従来、TC の進展に関わる危険因子はアルコールやたばことされてきたが、このような危険因子を有しない若い患者の例がかなり増えてきている。最近の研究により、ヒトパピローマウイルス (HPV) がそのような事例の病因として挙げられており、これは患者全体の一部における特有の危険因子となっている。HPV 陰性の TC に比べ、HPV 陽性の TC では p16 の過剰発現が見られ、EGFR の増幅は少ない。HPV-16 が感染した状態では c-myc の増幅と HIF-1a の過剰発現が見られる。HPV-16 の感染は、最初は扁桃陰窩において扁桃がんの進展に直接関係し、G1-S phase に関係した p16 の過剰発現のような細胞周期の撹乱がそれに続く可能性がある。
