A metallo-carboxypeptidase that is predominantly expressed as a membrane-bound enzyme of 94-100 kDa , but also exists in a soluble form. Hydrolyses alpha-peptide bonds in Ac-Asp-Glu, Asp-Glu, and Glu-Glu, but also gamma-glutamyl bonds in gamma-Glu-Glu, and folylpoly-gamma-glutamates. With folylpoly-gamma-glutamates, shows processive carboxypeptidase activity to produce pteroylmonoglutamate . Does not hydrolyse Ac-beta-Asp-Glu. Known inhibitors: quisqualic acid, Ac-beta-Asp-Glu, and 2-phosphonomethyl-pentanedioate. In peptidase family M28 of Vibrio leucyl aminopeptidase. The release of C-terminal glutamate from folylpoly-gamma-glutamates is also catalysed by EC 126.96.36.199 (glutamate carboxypeptidase) and EC 188.8.131.52 (gamma-Glu-X carboxypeptidase).
EC 184.108.40.206 created 1997, modified 2000 (EC 220.127.116.11 created 1972 and EC 18.104.22.168 created 1992, incorporated 2000)
